Amulet’s primary area of focus is in the area of gastrointestinal disorders including:

Amulet Pharmaceuticals, Inc is developing a completely new treatment for gastroparesis a complication of diabetes, which results in the “paralysis” of the stomach causing bloating and vomiting of food that does not move properly through the digestive tract. Our lead program, AMU-301, seeks to establish NORTECH™ Technology as the primary treatment for gastroparesis, a serious unmet medical condition through the precise delivery of nitric oxide to the affected tissues. Since the early 1990’s, the pivotal role of nitric oxide in gastroparesis has been established. It is now known that gastroparesis is caused by the death of the nitric oxide producing nerves of the stomach and GI tract. Amulet Pharmaceuticals, Inc AMU-301 replaces the lost nitric oxide by delivering biologically active nitric oxide directly to the stomach and gastrointestinal tract in a safe topical formulation.

There is a need for better therapies for gastroparesis. Current therapies are either ineffective or unsafe or both. New therapies in development may be effective and safe but can only be administered intravenously and are therefore only useful in emergency settings, or have the same mode of action as Cisapride, which was withdrawn by the FDA in 1999 for safety reasons. AMU-301 is the only potential therapy for gastroparesis currently in development that is safe and can be used as a prophylactic and given to more people for longer periods of time to reduce the overall impact of the disease.

All current treatments for gastroparesis act via systemic distribution, i.e. they are absorbed into the general circulation and act throughout the body, resulting in unwanted side effects. For example, metoclopramide acts in the GI tract to stimulate the stomach but also works on the brain to produce potentially serious side effects. Thus, these drugs treat the symptoms and not the root cause of the symptoms i.e. the loss of nitric oxide in the stomach. Amulet’s AMU-301 delivers nitric oxide topically in the stomach and cannot be distributed systemically.

AMU-301 can be used to treat other gastrointestinal conditions such as obesity, enteral feeding in critical care patients, irritable bowel syndrome, functional dyspepsia.
